I think a signature rule is the only way to get emails to come from a rep. You would only need to enter every rep into the rule once, but if you have over 1000 reps, you would need to make sure every rep's names included in the rule. There are several discussions here and many ideas submitted in the help portal to make the process for creating signature rules a lot less cumbersome.
